Does anyone have a definitive answer to whether or not the estimated number of recipients takes into account the lists selected as “Don’t send to” (aka the suppression lists)?

Yes, they do – or at least they’re designed to. If you want to test this yourself, simply create a list of all contacts (Create date is known) and exclude it in the Recipients tab. The count should go down to 0.
