If I want my first email to be a new conversation, then the second one to be a reply.
Then, my third one to be a new conversation with a new subject and then the fourth one to be a reply to the second email and so on.
Is this possible?

Are you referring to sequences? If so, then I’m not aware of any settings that would let you customize how exactly emails are threaded.
There is an option to thread emails as replies – this will create a single email thread here: Create an email thread in a sequence
