I am trying to configure Messenger Bot to not fire when someone has already messaged me.

Yes, it is possible to customize the targeting of your HubSpot chatbot so that it will exclude from contacts who have already messaged you before. You can find the settings under "Target" of your chatbot (screenshot #1 big).

Under 'Visitor information and behavior', you can add an exclusion rule using 'Segmented lists' as a filter (see screenshot attached). For the list, you can use an activity filter to segment contacts who have an associated activity with your chatflow. I have created one as an example (screenshot, smaller).
