We have a bunch leads that sign up to our site and potentially do not respond to our first email to book a meteing. If they do not respond I want to send an email 48 hours later via a worklow with a canned response.
Lifecycle stage at this point is subscriber and I would assume we would use the "no activity" within 48 hours trigger templated email?
Do you have the email address that they would send their response to connected to HubSpot? That would be the best first place to start.
If they reply to the email and do not book a meeting would you want them to enroll or only if they don't reply at all?
Is the email that gets sent out a 1:1 email or an automatic marketing email?
Also, how are you booking meetings? Do you use the HubSpot meetings tool?
Imagning that you do have the email connected and are using HubSpot meetings then the best way would be to use a contact-based workflow using activity triggers.
Once I know a little bit more about the initial email and how you are booking meetings i will be happy to advise you on the best path forward here.

(The main difference are the manual additional step + the fact that the above solution uses the connected marketing email domain while a sales email is sent from the contact owner's personal inbox.)
Without workflows, this process cannot be automated in HubSpot. You would have to create a list of contacts who submitted the form 2 days ago and for whom "Date of last meeting booked in meetings tool" is unknown. You could then use templates to work through this list of contacts.
