Hello Community,
Is there a way to update a Salesforce Lead status using a workflow, even thought the status property is setup to prefer Salesforce?

I don't think so. If you clear or change a 'prefer Salesforce' mapped property in HubSpot, the value in Salesforce will just overwrite it again.
I recommend you look for a workaround using, perhaps, different mapped properties and Salesforce Process Builder to either clear or set the Lead status on the Salesforce side.
