Has anyone successfully set up a way to receive a second notification if a form has not been responded to? I have tried creating the workflow but I am having trouble getting it to work.
@RPetrovsky this requires a Professional subscription. You would enroll contacts when they submit a form, delay for 1 day, the send an internal email notification. As a workflow goal, you would specify that 'Last contacted is less than 1 day ago'.
(Without workflows, this cannot be done.)

We receive forms from clients interested in speaking events. We want to reply in 24 hours and if we do not I would like our team to get a second notification (reminder) that the form has not been responded to.

Could you explain in more detail what you mean when you say "not responded to form"? To whom is that referring, who is (not) responding, to what, who should be notified?
