I have set a validation rule on my single line text field regarding           character limit but its not working at form level after publishing the form

Validation rules are not enforced in form submissions, see the yellow info box here: https://knowledge.hubspot.com/properties/set-validation-rules-for-properties

 

HubSpot will likely introduce this in the upcoming months (gut feeling) but as of now, it's not possible without custom development based on the forms API.
