I've done some digging, but unfortunately, I don't think you'll be able to add a nofollow only to one comment without being able to edit that comment directly in the database.
You could do something that made all comments nofollow, but it doesn't sound like that's ideal.
There may be other workarounds, but these are my initial conclusions.
One quick add...just based on my opinion, if the link isn't worthy of a follow, the comment may not be worth keeping.
