I am designing an automation solution where I may need to bulk reassign a contact's contact owner (who will be an employee) if that employee leaves a company. What series of API calls would be the best approach to this solution?

Since HubSpot Users (required for contact ownership) cannot be created via the API, AND since Contact.Owner is easily changed (in bulk) via the UI, the solution here is probably best performed via the UI as opposed to performing via an integration. In other words, no integration steps can be initiated to change contact ownership to a HubSpot User until after that user has been created via the UI first.

A user does not need to be created in our solution. They will already exist as a Hubspot User. When the employee leaves the company, we want to integrate a system to update/change the contact owner for any contact in which the contact owner was the former employee. Hope this helps.
